Sampling Methods

多在步道或道路旁20公尺範圍內採集

Study Extent 研究範圍:臺灣
Quality Control 標本館內之標本採集者眾多,也有一般民眾捐贈之標本,以下步驟為特生中心目前的採集、鑑定標準流程。

Method step description:

  1. 標本採集方法 (一)保持標本的完整,以採集刀刮下或剝下,樹枝或葉附生者以修枝剪處理,若採集之標本數量足夠,標本面積以手掌心之大小為原則,若數量稀少,將1–3株植物體帶回鑑定即可。 (二)土生及石生的標本,盡量將其基質清除,留在原地,減少將土壤中的生物帶回標本館,也減少標本的重量;樹生的標本需注意採集時不要傷到樹皮,盡可能地小心將標本與基質分離;若標本的水分很多,可輕握於手中,將水分排除。 (三)標本採集後置於紙袋中,紙袋建議使用80磅–120磅之牛皮紙,苔蘚植物能吸收相當多的水分,使用牛皮紙能避免紙袋吸收水分而破裂,地衣較為乾燥,但若於雨天採集,一樣會有相當多的水氣。 (四)紙袋可將光度、濕度、生態環境、生長基質等各項基本資料事先印刷,採集時依紙袋上選項內容圈選,並視需要註記相關事項後,將標本置於保鮮盒中,保鮮盒可存放多份標本避免紙袋摩擦破裂並保持水分。 (五)回到實驗室後,依需要可以相機補拍微距相片,或將標本置於保鮮盒中放入冰箱保存,以進一步記錄其活體細胞特徵,惟需注意溼度控制,避免乾燥死亡或過濕腐爛發霉,依經驗以在2週內處理完畢為宜。 (六)若不進行前述處理,可直接將標本紙袋的標本排置於多孔塑膠籃中,以電風扇風乾或置於40℃以下之循環烘箱2–3天,即可得到品質良好的苔蘚標本。
  2. 標本鑑定方法 (一)新鮮野外標本,在未鑑定前須先保濕於生長盒中,置於冰箱中培養保存。各標本館借閱之乾燥標本於檢視前則需浸泡於清水或2% KOH水溶液中膨潤恢復原形。 (二)將標本裁切解剖後,製作成玻片,於實驗室中以解剖及光學顯微鏡,配置繪圖器,作形態分類之描述、量度,並加以照相及繪圖。 (三)使用各相關專論、專書及研究報告,配合模式標本及權威標本之比對,進行標本鑑定工作。
