Description
了解生物多樣性為研究生態及演化議題之基本條件。我們可將生存於極端環境下的溫泉紅藻(Cyanidiales) 視為模式生物,藉由它們來了解物種多樣化及種化過程中在生態學上及遺傳的機制。本研究利用選殖技術定序技術進行環境基因體rbcL基因之研究,調查臺灣火山區溫泉紅藻生物多樣性。研究結果顯示,目前臺灣溫泉紅藻可被分為Galdieria maxima, Galdieria partita及Galdieria sulphuraria三個物種。

Sampling Methods
採樣時,針對岩石及土壤樣本,以榔頭與金屬鏟子於含有溫泉紅藻群落的石塊或泥土進行採樣;針對溫泉水中樣本,以牙刷刷取含有溫泉紅藻的石塊,並將樣本收集於50毫升(ml)離心管中

Method step description:
- 採集樣品時,使用榔頭跟金屬鏟子於含有溫泉紅藻群落的石塊或泥土進行採樣,每一次採樣所含樣本面積大小約為 4 平方公分,樣本保存於高溫保溫盒中,待運回實驗室再做進一步處理。回到實驗室後,將保存於高溫保溫盒的樣品取出,並做兩部分處理,一部分保存於 -80˚C冰箱以利於之後的細胞核甘酸萃取;另外一部份利用培養基稀釋純化,進行單一藻種分離並大量培養,以利後續實驗分析。
